Abstract
Storage conditions were shown to affect the viability and moist heat resistance of Bacillus stearothermophilus spore suspensions. Refrigerated storage (4 °C) in distilled water did not significantly affect spore viability, but had an adverse effect on moist heat resistance. Freezer storage (–10 °C) and the subsequent thawings and refreezing of the suspensions was shown to have a significant effect on spore viability without a concomitant loss of moist heat resistance in the surviving population. Refrigerated storage (4 °C) in 0.02 M calcium acetate, pH 9.7, stabilized spore resistance without a significant loss of viability for a minimum of two years.
